SNARE complex (SNAP-25 competitive binding site)
Pathway
hCAP-18 precursor → Proteinase-3 cleavage → LL-37 release → Membrane insertion/disruption
Acetyl octapeptide-3 → SNARE complex disruption → Reduced vesicular fusion → Decreased acetylcholine release → Muscle relaxation
Downstream effect
Membrane permeabilization, cytokine induction, autophagy, phagosome-lysosome fusion, chemotaxisAhmad 2026Zhang 2026
Transient reduction in neuromuscular signal transmission, decreased muscle contraction amplitude, wrinkle depth reduction

N/A — topical cosmetic, no systemic endocrine axis
Origin
Endogenous human cathelicidin (37-AA fragment, residues 134–170 of hCAP-18)
Synthetic peptide derived from N-terminal fragment of SNAP-25 protein (synaptosomal-associated protein 25 kDa)

Parameter
LL-37
SNAP-8
Cytotoxicity (high dose)
Membrane disruption in host cells at supraphysiological concentrations
Pro-inflammatory signaling
Can exacerbate inflammation in certain contexts (context-dependent)Pinheiro 2026
Biofilm formation risk
LL-37-DNA complexes may stabilize dental plaque biofilmsTanabe 2026
Theoretical cancer risk
Immunomodulatory roles in tumor microenvironment under investigation
Cytotoxicity
Concentration-dependent antiproliferative effect observed in vitro; IC50 ~10 mg/mL (argireline, 6-AA analogue)

Hypersensitivity
Rare; no widespread allergic reactions reported
Absolute Contraindications
LL-37
SNAP-8
  • ·Known hypersensitivity to acetyl octapeptide-3 or formulation excipients
Relative Contraindications
LL-37
  • ·Active autoimmune disease (theoretical immune dysregulation)
SNAP-8
  • ·Active skin infections or open wounds at application site
  • ·Neuromuscular disorders (theoretical concern, no documented cases)

05Administration Protocol

Parameter
LL-37
SNAP-8
1. Natural secretion
LL-37 is constitutively expressed in neutrophils and epithelial cells, cleaved from hCAP-18 by proteinase-3 at sites of infection or inflammation.
Wash face with gentle cleanser and pat dry. Remove makeup and surface oils to optimize peptide contact.
2. Experimental formulations
Synthetic LL-37 and derivatives (e.g., SAMP-12aa) tested in vitro and animal models. Administered via topical, intraperitoneal, or intravenous routes in research settings.
Apply 1–2 drops or pea-sized amount of SNAP-8 serum or cream to target areas (forehead, glabellar lines, crow's feet). Gently massage until absorbed.
3. Stability considerations
LL-37 is resistant to pepsin degradation at gastric pH. Synthetic short peptides designed to retain this stability while reducing toxicity.Lu 2026
Twice daily — morning and evening. Allow 2–3 minutes for absorption before applying additional skincare products.
4. Layering
Apply before heavier creams or occlusive moisturizers. Peptides penetrate best from water-based serums on clean skin.
5. Storage
Store at room temperature, away from direct sunlight. Refrigeration may extend shelf life of formulations containing unstable peptides.
6. Duration
Consistent use for 20–60 days required for visible wrinkle reduction. Effects are temporary and reverse upon discontinuation.
